## Key facts

- In a preprint, untreated pancreatic cancer cells showed a reproducible bias toward cell-cycle-arrest and anti-apoptotic programmes. (self-reported) [cites: 1793977388404318473]
- In a preprint, TIF1g-deficient CD8+ T cells proliferated more but expressed less Granzyme B and Perforin, and loss of TIF1g accelerated B16 melanoma growth in mice. (self-reported) [cites: 1793977388404318477]
- In a preprint, the REV1 inhibitor JH-RE-06 showed single-agent growth-inhibitory activity in patient-derived colorectal cancer organoids. (self-reported) [cites: 1793977388404318472]
- In a preprint in a mouse model, the fibronectin receptor Embigin was upregulated in premalignant and cancerous gastric tissue and its expressing cells co-expressed the cancer stem cell marker CD133. (self-reported) [cites: 1793977388404318478]

## What changed

Four preprints posted this week describe cancer as a problem of state, not just of growth. In untreated pancreatic cancer, malignant cells lean toward pausing rather than dying. In the stomach, a marker of tumour development persists from premalignancy all the way to cancer. In the immune system, T cells that lose a key regulator multiply enthusiastically but cannot kill. And in colorectal cancer, a drug target only helps in some contexts, and works through mechanisms nobody expected. The shared lesson is that whether a cell divides, dies or waits depends on programmes that can be read, and in principle shifted. [cites: 1793977388404318473, 1793977388404318478, 1793977388404318477, 1793977388404318472]

That framing matters because most cancer drugs are judged by whether tumours shrink, which is a test of killing. These papers suggest the more informative question is often what state the cells are in before treatment starts, and whether the treatment changes that state. A tumour full of arrested cells, an immune response full of busy but ineffective T cells, and a drug that works in one cell line and not another are all versions of the same observation: the context decides the outcome. [cites: 1793977388404318473, 1793977388404318477, 1793977388404318472]

## The pause in pancreatic cancer

The pancreatic cancer preprint studied untreated pancreatic ductal adenocarcinoma and found a reproducible transcriptional bias toward anti-apoptotic and cell-cycle-arrest programmes. TGM2 expression was modestly higher on the arrest-associated side in both cohorts, making it a candidate marker of that state. [cites: 1793977388404318473]

The honest limit is stated in the paper itself: the contrast between arrest and apoptosis programmes did not track cell-line drug sensitivity or survival in bulk tumours. So the arrest bias is a baseline description of the untreated tumour's intrinsic state, not a predictor of who will respond to treatment. That is still useful, because any claim about how a drug changes pancreatic cancer has to be measured against what the tumour looked like before treatment, and this paper supplies that baseline in unusual detail. It also cautions against reading a paused tumour as a defeated one. [cites: 1793977388404318473]

## T cells that multiply but cannot kill

The immunology preprint concerns TIF1g, also known as TRIM33, a regulator that couples TGF-b signalling to CD8+ T cell activation and cytotoxic function. When TIF1g was missing, CD8+ T cells bypassed the usual TGF-b suppressive signals and became hyper-activated and hyper-proliferative, but they failed to express enough Granzyme B and Perforin, the molecules that do the killing. In living mice, loss of TIF1g accelerated growth of B16 melanoma tumours, with reduced Perforin expression in the tumour-infiltrating CD8+ T cells. [cites: 1793977388404318477]

This suggests TIF1g acts as a threshold regulator that balances proliferative expansion against cytotoxic effector differentiation, keeping the two programmes coupled so that more T cells also means more killing. Without it, expansion and killing come uncoupled: plenty of cells, little effect. The researchers present TIF1g as a potential target for enhancing T cell-based immunotherapies, but the work is in mice, with no clinical validation in human patient cohorts, and the mechanism of how TIF1g interacts with TGF-b downstream effectors still needs structural characterisation. The practical warning is sharper than the therapeutic promise: counting T cells is not the same as measuring an immune response. [cites: 1793977388404318477]

## A context-dependent drug, and a marker that persists

The colorectal cancer preprint tested REV1 as a drug target. The inhibitor JH-RE-06 enhanced chemotherapy sensitivity in colorectal cancer cell lines, but only in a context-dependent manner, and it also showed single-agent growth-inhibitory activity in patient-derived colorectal cancer organoids. The mechanism was not the simple DNA-damage story the target implies: REV1 inhibition induced accumulation of LC3-II and reduced beta-tubulin, suggesting autophagy and cytoskeletal involvement. [cites: 1793977388404318472]

The gastric cancer preprint traced a tumour's development from premalignancy to cancer in CTLA4-knockdown mice. It identified a metaplastic population derived from chief cells that serves as a precursor to gastric adenocarcinoma, with fibronectin signalling increasing progressively from premalignancy to cancer. Embigin, a fibronectin receptor, was upregulated in premalignant and cancerous gastric tissue and persisted through progression, and Embigin-expressing epithelial cells co-expressed the cancer stem cell marker CD133. There are no clinical trial data and no functional inhibition studies of Embigin in living animals, so it is a candidate marker and target for early detection rather than a proven one. [cites: 1793977388404318478]

## What is still unresolved

- All four papers are preprints, not yet peer reviewed.
- The pancreatic arrest-apoptosis contrast does not predict bulk survival or drug sensitivity.
- The TIF1g work has no clinical validation in human patient cohorts.
- REV1 inhibition has no clinical validation, and the mechanism of beta-tubulin reduction is not fully elucidated.
- Embigin has no clinical trial data and no functional inhibition studies in vivo.

## Decision card

- **Useful for:** Cancer researchers studying tumour cell state before treatment; Immunotherapy developers measuring T cell function rather than T cell counts; Drug developers weighing REV1 or TIF1g as targets
- **Not for:** Clinical decisions, which none of these preprints supports; Treating Embigin or TGM2 as validated clinical biomarkers; Assuming a drug that works in one cell line or organoid will work in another
- **Compare against:** Tumour shrinkage as the sole measure of drug effect; T cell counts as a measure of immune response; Single-context drug sensitivity testing
