## Key facts

- In a preprint, an engineered yeast strain yields 4.05 mg of a dual GLP-1/GIP precursor per gram of dry biomass, but a 12-week trial in 98 adults with overweight found no statistically significant weight-loss difference versus dietary guidance. (self-reported) [cites: 1792859290845257235]
- In a preprint, T cells carrying a G2R chimeric receptor expand in response to G-CSF and induce complete, durable regression in mouse melanoma models without lymphodepletion or added IL-2. (self-reported) [cites: 1792858925570870375]
- In a preprint, mucosal cytokines drive B cells into antibody-producing cells in IgA nephropathy patients, and the resulting pathogenic antibody populations are heavily lambda-enriched compared with controls. (self-reported) [cites: 1792858925570567481]

## A yeast that delivers a hormone — and a null result

The most instructive of the three preprints is the one that did not work as hoped. An engineered yeast strain yields 4.05 milligrams of pro-DALAP, a dual GLP-1/GIP receptor agonist precursor, per gram of dry biomass — a living factory for a gut hormone, deliverable by mouth rather than by injection. [cites: 1792859290845257235]

But the human test disappointed: a 12-week randomized trial of pro-DALAP in 98 adults with overweight showed no statistically significant difference in weight loss compared with dietary guidance. Preliminary rat toxicology studies, at 300 micrograms per kilogram per day for 7 days, showed no treatment-related adverse findings, so safety is not the reported problem — efficacy, at this dose and design, is. Publishing a clean null like this is what keeps the field honest about oral biologics. [cites: 1792859290845257235]

The platform idea is what makes the null result worth reporting rather than burying: if yeast can manufacture and deliver a gut hormone by mouth, the same approach could in principle carry other biologics that today require injection. The trial says this version, at this design, does not move weight — which is exactly the information the next version needs. [cites: 1792859290845257235]

## T cells switched on by a growth factor

The second preprint shows the control idea working, so far only in mice. A G2R chimeric receptor enables human T cells to expand in response to G-CSF, a growth factor, rather than relying on the interleukin signals that normally drive T cell growth. [cites: 1792858925570870375]

In B16-F10 melanoma models, G2R-expressing T cells induce complete, durable regression without lymphodepletion or exogenous IL-2 — two of the harshest parts of conventional cell therapy are sidestepped. And the control persists: G2R-expressing memory T cells are capable of re-expansion upon secondary G-CSF treatment, enabling rejection of secondary tumor challenges. The therapy is a switch as much as a weapon. [cites: 1792858925570870375]

Avoiding lymphodepletion matters because it is one of the most punishing parts of cell therapy: wiping out a patient's existing immune cells to make room for the engineered ones. A receptor that lets growth factor alone drive expansion, and re-expansion on demand, points toward cell therapies that are gentler to receive and easier to re-dose. [cites: 1792858925570870375]

## A kidney disease driven from the mucosa

The third preprint shows the same signal logic from the disease side. In IgA nephropathy patients, mucosal cytokine stimulation — with IL-6, IL-21, BAFF, and APRIL — induces robust differentiation of B cells into antibody-producing plasmablasts and plasma cells. [cites: 1792858925570567481]

The resulting pathogenic antibody populations are disproportionate: patients demonstrate an expansion of galactose-deficient IgA1 populations enriched for lambda light chains at about 90%, compared with about 51% in healthy controls. Mucosal cytokine signals directly amplify pathogenic B cell responses in IgA nephropathy, providing mechanistic validation for gut-directed corticosteroids and BAFF/APRIL inhibition. Where the yeast and T cell preprints engineer signals, this one maps a disease that already runs on them — and points treatment at the signal source. [cites: 1792858925570567481]

Read together, the three preprints describe a field learning to steer biology rather than simply dose it — and learning, in the yeast trial's case, that steering is not the same as succeeding. All three are preprints, and the T cell work is in mice, so none of this is a treatment yet. But the direction is consistent: control the signal, and the therapy follows. [cites: 1792859290845257235, 1792858925570870375, 1792858925570567481]

The therapeutic pointer is concrete rather than speculative: because mucosal cytokine signals directly amplify the pathogenic B cell response, the preprint provides mechanistic validation for gut-directed corticosteroids and BAFF/APRIL inhibition — treatments aimed at the signal source in the mucosa rather than at the kidney damage downstream. [cites: 1792858925570567481]

## What is still unresolved

- Whether the yeast platform works at a different dose or formulation
- Whether G-CSF-controlled T cells translate from mouse melanoma to human tumors
- Whether BAFF/APRIL inhibition changes IgA nephropathy outcomes in trials

## Decision card

- **Useful for:** Tracking engineered living therapies; Reading early-stage therapy evidence critically
- **Not for:** Treatment decisions; Assuming mouse or early human results transfer to patients
- **Compare against:** Injectable GLP-1 drugs; Conventional CAR-T and cytokine therapies
